Real Time PCR Systems Instructions |
The Applied Biosystems 7500 Real Time PCR Systems uses fluorescent-based PCR
chemistries to provide quantitative detection of nucleic acid sequences using
real-time analysis and qualitative detection of nucleic acid sequences using
end-point and association-curve analysis.
ABI Real Time PCR 7500 Chemistry: Applied Biosystems has developed two types
of chemistries that can be used to detect PCR products using 7500 Real Time PCR
Systems:
1) TaqMan Probe-based
fluorogenic 5' nuclease chemistry.
2) SYBR Green I dye
chemistry.
Applications:
ABI7500 Real Time PCR Systems
can be used for:
1)
Absolute Quantification: to determine the absolute quality of a
single nucleic acid target sequence within an unknown sample.
2)
Relative Quantification: to determine the change in expression of a
nucleic acid sequence (target) in a test sample relative to the same sequence in
a calibrator sample.
3)
Plus/Minus Assay: to determine if a specific target sequence is
present (plus) or not present (minus) in a sample.
4)
Allelic Discrimination : to detect variants of a single nucleic acid
sequence, SNP.

Materials:
For using ABI 7500 Real Time
PCR Systems, users need to provide their own 96 well plates (Applied Biosystems
PN 4306757), Optical Adhesive Cover (Applied Biosystems PN 4311971),
TaqMan RT-PCR reagents,
TaqMan Universal PCR Master Mix (Applied Biosystems PN 4304437),
TagMan Probe based probe or primers or
SYBR Green based reagents, also tubes and tips for PCR reaction (for details
please consult your specific application protocol or visit
Applied Biosystems homepage).
