Creating an Absolute Quantification (AQ) Plate Document :
Log on to 7500 computer with your own user ID and password (will be created for you after you fill out the order form)
1. Select Start>Programs>Applied Biosystems7300/7500>Applied Biosystems 7300/7500 SDS Software to start the SDS (Sequence Detection Software).
2. Select File>New.
3. In the Assay drop-down list of the New Document Wizard, select Absolute Quantification (Standdard Curve). Accept the default settings for Container and Template (96-well Clear and Blank Document).
4. Enter a name in the Default Plate Name field or accept the default.
5. Click Next>.
6. Select detectors to add to the plate document.
a. click to select a detector.
b. click Add>>. The detectors are added to the plate document.
c. click Next>.
7. Specify the detectors and tasks for each well.
a. click on a well to select it
b. click on the detector name to select the detector for the well.
c. click under the Task column to assing the detector task.
d. Enter a quantity for wells that contain standards
e. click Use.
f. click Finish.
8. Enter the sample names.
a. select View>Well Inspector
b. click on a well or click-drag to select replicate wells.
d. if necessary, change the settings for the Passive Reference dye, or ROX will be used.
e. repeat steps b through d until you have specified sample names and passive reference dyes for all the wells on the plate.
f. click (x) to close the Well Inspector.
9. Verify the information on each well in the Setup tab.
