Specifying Thermal Cycling Conditions and Starting the Run:

There is a default thermal cycling condition for the PCR step of the procedure:

1. Select the Instrument tab

2. Verify that:

Sample volume is 50 ul

9600 Emulation is selected.

if you are using SYBR Green I chemistry, click Add Dissociation Stage.

3. Select File>Save As, enter a name for the AQ Plate document, then click Save.

4. Load the plate into the instrument.

5. Click Start.

After the run, click Analysis buttun.

2. Adjusting the Baseline and Threshold

3. Analyzing and Viewing the AQ Data

4. Exporting AQ Plate Data
