ANALYZING AQ DATA
Configuring Analysis Settings:
1. click Analysis button
2. in the Detectors drop-down list, select All.
3. select Auto Ct. The SDS will automatically generate baseline and threshold value for each well.
4. select Use System Calibrator if you want to use the calibration files.
5. click OK & Reanalyze.
6. examine the amplification plot, and if necessary manually adjust the baseline and threshold.
Adjusting the Baseline and Threshold:
A typical amplification curve has a:
Plateau phase
Linear phase
Geometric phase
Background
Baseline
Analyzing and Viewing the AQ Data:
click Results tab to see:
Plate Tab: showing the plate setting.
Spectra Tab: showing the fluorescence spectra of selected wells.
Component Tab: Showing the complete spectral contribution of each dye in a selected well over the duration of the PCR run.
Amplificatioin Plot Tab: showing: 1) Rn vs. Cycle (Linear) View; 2) Rn vs. Cycle (Log) View: 3) Ct vs. Well Position View 4) Standard Curve; 5) Dissociation; 6) Report.
Exporting AQ Plate Data
1. Select File>Export
2. Enter a file name for the export file
3. Click Save
